Inhibition of thrombin and FXa generation by curcuminand BDMC in HUVECs. (A) FVa (100 pM) and FXa (1 nM) werepreincubated on HUVECs monolayers for 10 min with indicatedconcentrations of curcumin (□) or BCMC (■). Prothrombin wasadded to a final concentration of 1 μM and the amounts of prothrombinactivated were determined at 30 min as described in“Materials and Methods”. (B) HUVECs were preincubated with indicatedconcentrations of curcumin (□) or BCMC (■) for 10 min.TNF-α (10 ng/ml for 6 h) stimulated HUVECs were incubated withFVIIa (10 nM) and FX (175 nM) in the absence or presence ofanti-TF IgG (25 μg/ml) and FXa generation was then determinedas described in “Materials and Methods”. (C) HUVECs were culturedwith curcumin (□) or BCMC (■) in the absence or presenceof TNF-α (10 ng/ml) for 18 h and PAI-1 concentration in theculture mediums was examined as described in “Materials andMethods”. (D) the same as C except that cells were preincubatedwith curcumin (□) or BCMC (■) and cultured with SP600125 (2μM), emodin (2 μg/ml), and PD98059 (10 μM) in presence ofTNF-α (10 ng/ml) for 18 h. *P < 0.05 as compared to 0 (A) orTNF-α alone (B, C) or to (+) control (D).
