Agar well diffusion Tubes of 20 ml sterile liquid nutrient agar (OXOID) at 50◦Cwere inoculated with 500 l of 7 log CFU/ml Staphylococcus aureus (overnight cultures grown at 37◦C on nutrient broth, and plated out in nutrient agar media to take colony count), mixed thoroughly ,poured into sterile empty plates and left 1 hour at room temperature until solidification.

วุ้นดีแพร่ของ 20 มล.ผ่านการฆ่าเชื้อของเหลวสารอาหารวุ้น (OXOID) ที่ 50◦Cwere inoculated กับ l 500 ของล็อก 7 CFU/ml หมอเทศข้างลาย Staphylococcus (ข้ามวัฒนธรรมปลูกที่ 37◦C บนสารอาหารซุป และชุบสื่อสารอาหารวุ้นจะใช้จำนวนอาณานิคม), ผสมอย่าง เทลงในจานเปล่าผ่านการฆ่าเชื้อ และหลอดเหลือ 1 ชั่วโมงที่อุณหภูมิห้องจนกระทั่งแข็งตัว
