DNA sequence analysis showed all DNA clones (ten for each three
spots) had identical sequences of their 18S rRNA gene. BLAST search
showed that the generated sequence (1667 nucleotides) was highly
similar to members of the Sphaeropleales (Chlorophyceae). In more
detail, it showed highestmatch (99.7% similarity) with Desmodesmus
armatus var. subalternans (Smith) Hegewald [formerly Scenedesmus
armatus var. subalternans Smith] and a few other Desmodesmus
species (more than 99% similarity) while clearly separated from a
sister genus, Scenedesmus (ca. 96% similarity, see Fig. 3). Further
DNA analysis based on ITS2 region (including partial 5.8S rRNA
gene; total 352 nucleotides) showed all DNA clones again exhibited
identical sequence and 100% matchwith that of D. komarekii (Hegewald)
Hegewald.
