The effects of enzyme inhibitors on

The effects of enzyme inhibitors on protease and -
amylase activities were studied using phenylmethylsulfonyl
fluoride (PMSF), ethylenediaminetetraacetic acid (EDTA), -
mercaptoethanol and dithio-bis-nitrobenzoic acid (DTNB). The
NH1 crude enzyme was pre-incubated with inhibitors for 30min
at 25 ◦C, and then the remaining protease and amylase activities
were determined under standard assay conditions. The activity of
the crude enzyme assayed in the absence of inhibitors was taken as
100%.
The effect of various metal ions (5mM) on protease and amylase
activitieswas investigated by adding divalent metal ions (Ca2+,
Mn2+, Zn2+, Cu2+, Ba2+, Mg2+) to the reaction mixture. The activity
of the crude enzyme without metallic ions was considered as 100%.
