The combination of polytene cytology with the genet- ics of Drosophila has proven to be a powerful approach in the dissection of chromosomal events underlying tran- scription. Combining an allelic series of Ash 1 mutations with immunostaining for dimethylated lysine 4 of histone H3 (H3K4), Byrd and Shearn [7] demonstrated the requirement for ASH1 in dimethylation of H3K4, a chro- matin mark for gene activation.
